Scientists at the Jawaharlal Nehru Centre for Advanced Scientific Research (JNCASR), an autonomous institute of the Department of Science and Technology, have developed TZ-48, a fluorescence lifetime-responsive small-molecule probe that detects amyloid-beta (Abeta), the earliest and most Alzheimer's-specific biomarker of the disease.
The team paired the probe with ADxFluor, a smartphone-integrated platform that reads TZ-48's change in fluorescence intensity and quantifies the Abeta load in blood serum, turning what today needs a PET or MRI suite into a potential point-of-care test.
TZ-48 crosses the blood-brain barrier, labels Abeta plaques in transgenic Alzheimer's mouse brains, works with confocal laser scanning microscopy and fluorescence lifetime imaging microscopy, and detected Abeta in cerebrospinal fluid and serum accurately enough to separate diseased from healthy mice; the work is published in ACS Chemical Neuroscience.
Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by memory loss, cognitive decline and behavioural impairment. Central to its pathology is the misfolding and aggregation of the amyloid-beta peptide, which self-assembles into fibrils and is deposited outside neurons as extracellular plaques; these set off neurotoxic cascades that damage synapses and kill neurons. The second hallmark is the tau protein, which normally stabilises the internal transport skeleton of a neuron but in AD becomes abnormally modified and twists into neurofibrillary tangles inside the cell. The framework of the National Institute on Aging and the Alzheimer's Association (NIA-AA) - the US reference standard for defining AD biologically - groups the diagnostic markers into three: amyloid (A), tau (T) and neurodegeneration or neuronal injury (N), the widely cited AT(N) system, measurable in brain imaging, CSF and blood. Abeta pathology appears early and is specific to Alzheimer's, which is why a cheap Abeta test is the prize. Alzheimer's is the commonest form of dementia, but dementia is the broader umbrella - vascular dementia, Lewy body dementia and frontotemporal dementia are separate conditions.
Simple Analogy: Amyloid plaques are like sticky debris piling up between houses on a street, while tau tangles are the collapsed internal scaffolding inside each house. TZ-48 is a dye that lights up only when it touches that particular debris - and a phone camera reads how brightly it glows.
A fluorescent molecule absorbs light at one wavelength and re-emits it at a longer one. Two properties can be measured: intensity (how bright the glow is) and lifetime (how long the molecule stays in the excited state before emitting). Fluorescence lifetime imaging microscopy exploits the second, and this is the technical point of the JNCASR work: lifetime is largely independent of how much probe is present and of photobleaching (the fading of a dye under prolonged illumination), so it gives a robust, quantitative reading rather than a brightness that varies with dose. TZ-48 gives a 'turn-on' response - it is dim on its own and lights up on binding Abeta fibrils - and its distinct lifetime signatures allow differential detection across disease stages. The probe also shows blood-brain barrier (BBB) permeability. The BBB is the tight lining of the brain's capillaries, formed by endothelial cells joined by tight junctions with astrocyte and pericyte support, which blocks most large or water-soluble molecules from entering brain tissue; it protects the brain but is the single biggest obstacle in neurological drug and probe design, and a molecule that crosses it is therefore valuable well beyond this one application.
Simple Analogy: Measuring intensity is like judging a torch by how bright it looks - it depends on how close you hold it. Measuring lifetime is like timing how long the bulb takes to go dark after you switch it off: the same answer however many torches you have.

Its Bioorganic Chemistry Laboratory, which produced TZ-48, works on Alzheimer's amyloids, molecular probes and biomaterials, and earlier produced the candidate anti-amyloid molecule TGR63.

Together they issued the NIA-AA Research Framework, which defines Alzheimer's biologically by biomarkers rather than symptoms and gives the AT(N) classification cited in this release.

Tests performed at or near the patient rather than in a central laboratory - the same design logic behind rapid antigen kits and portable glucometers. The examinable trade-off is between laboratory-grade accuracy and reach: a screening test that costs little and needs no specialist can be deployed at a primary health centre, which is exactly where India's dementia burden goes undetected.
