Indian researchers have developed RK-251, a smart cancer drug that stays inactive in healthy tissue and switches on inside tumour cells.
It was developed jointly by Dr Asis Bala of the Institute of Advanced Study in Science and Technology, Guwahati, and Dr K.P. Bhabak of IIT Guwahati.
RK-251 is a prodrug triggered by reactive oxygen species, which are present at higher levels inside many cancer cells.
On activation it releases NBDHEX, an anticancer compound that blocks proteins cancer cells rely on to survive.
In preclinical tests it acted on triple-negative breast cancer cells while sparing healthy cells; zebrafish embryo studies showed no obvious toxicity.
A prodrug is a compound that is administered in an inactive or less active form and becomes active only after a chemical or metabolic trigger inside the body. Prodrugs are used to improve absorption, to get a molecule past a barrier it could not otherwise cross, or, as here, to confine a drug's action to the tissue where the trigger is present. Levodopa is a classic example: it crosses the blood-brain barrier, which dopamine itself cannot, and is converted to dopamine in the brain.
Simple Analogy: It is a locked package that only the destination has the key to open, so nothing spills out along the way.
