Researchers led by Dr. Asis Bala of the Institute of Advanced Study in Science and Technology (IASST), Guwahati — an autonomous institute of the Department of Science & Technology — and Dr. K.P. Bhabak of IIT Guwahati have developed a 'smart' anticancer molecule named RK-251.
RK-251 stays inactive as it travels through the body and is switched on by the abnormally high levels of reactive oxygen species (ROS) inside cancer cells, where it releases the anticancer compound NBDHEX; NBDHEX blocks target proteins that cancer cells use to survive and resist treatment.
In preclinical work the molecule showed strong activity against aggressive triple-negative breast cancer cells with much less effect on healthy cells, and no obvious toxicity in zebrafish (Danio rerio) embryos; the work is published in the ACS Journal of Medicinal Chemistry and further studies are needed before human testing.
A prodrug is a chemically masked, biologically inactive form of a medicine. It circulates harmlessly until some condition specific to the target tissue removes the mask and liberates the active drug. RK-251 uses reactive oxygen species (ROS) as that condition. ROS are chemically aggressive oxygen-containing molecules produced during normal cell metabolism; cancer cells, because of their distorted metabolism, generate them at far higher levels than normal cells. A chemical group that is cleaved by ROS therefore behaves as a lock that only tumour cells can pick. When the lock opens inside the cancer cell, RK-251 releases NBDHEX, a nitrobenzoxadiazole-based inhibitor of the enzyme glutathione transferase P1-1 (GSTP1-1) — an enzyme tumour cells lean on to detoxify drugs and to keep survival signalling alive. The same chemistry also makes the molecule fluoresce when ROS is present, which is how the researchers could see activation in the zebrafish embryos.
Simple Analogy: Conventional chemotherapy is a weedkiller sprayed over the whole garden — it kills the weeds and scorches the flowers. A ROS-activated prodrug is a capsule of weedkiller that only dissolves in the peculiar soil chemistry under a weed.

TNBC is breast cancer whose cells lack oestrogen and progesterone receptors and do not over-express HER2. Because the three receptors that hormone therapy and HER2-targeted drugs act on are absent, chemotherapy has been the main systemic option — which is exactly why a selective, ROS-activated molecule is being tested against it. TNBC accounts for roughly 15% of invasive breast cancers.
Danio rerio embryos are transparent, develop outside the mother and grow fast, so a fluorescent compound can literally be watched working inside a living animal. This is why zebrafish sit between cell-culture work and mammalian trials in the preclinical pipeline.
IASST is the DST's principal research anchor in the North East, created explicitly to work on the region's problems. Research output of this kind is the counter-argument to the perception that frontier science in India is confined to the metros.
